Paul Wesley föddes under namnet Paul Thomas Wasilewski i New Brunswick, New Jersey och han växte upp i Marlboro, New Jersey. Efternamnet Wasilewski kommer från hans föräldrar Agnieszka och Thomas som flyttade från Polen till USA strax innan Paul föddes. Det nuvarande efternamnet tog Paul 2005 eftersom Wasilewski var för svårt att uttala, och han säger att det har hjälpt hans karriär. Under uppväxten spenderade Paul varje år fyra månader i Polen, vilket har lett till att han idag pratar flytande polska. Han har en äldre syster, Monika och två yngre systrar, Julia och Leah.

Skoltiden spenderade Paul på Christian Brothers Academy och Marlboro Highschool, båda i New Jersey. Under sitt andra år i highschool fick Paul en roll i serien The Guiding Light som Max Nickerson. Detta ledde till att han bytte skola eftersom det blev för långt att pendla mellan skola, hem och jobb. År 2000 gick Paul ut highschool och började college på Rutgers University. Men där gick han bara en termin då han med sina föräldrars stöd valde att hoppa av och satsa på skådespelarkarriären istället.

Sedan dess har han haft roller i ett flertal serier som 24, 8 simple rules, American Dreams, Army Wives, Cane, Everwood, Guiding Light, Smallville, O.C och Wolf Lake.

Paul gillar att spela ishockey och åka snowboard. Han har också ett stort intresse av att regissera och skriva manus.
